Takeru Tenkūji (diperankan oleh Shun Nishime) seorang pemuda yang cheerful dan ceria. Mempunyai seorang ayah yang merupakan seorang pemburu hantu yang meninggal sekitar 10 tahun yang lalu. Suatu hari, Takeru dibunuh dan meninggal di tangan seorang monster jahat yang dikenal sebagai Ganma (眼 魔, arti dari "Eye Demon") ketika dia mencoba untuk melindungi teman masa kecilnya Akari Tsukimura (diperankan oleh Hikaru Ohsawa) dari serangan Ganma. Seorang pertapa misterius muncul dan membawa Takeru kembali ke kehidupan di dunia nyata dan memberikan kepadanya sebuah sabuk transformasi bernama Ghost Driver dan Eye-cons (眼 魂 (ア イ コ ン) Aikon arti "Souls Eye"), sebuah perangkat yang dapat melihat makhluk-makhluk hantu dan roh-roh makhluk yang sudah mati seperti Ganma dan juga dapat membuat Takeru untuk bisa berubah menjadi Kamen Rider Ghost. Dalam perjalanan cerita, muncul seorang pemuda misterius bernama Makoto Fukami (diperankan oleh Ryosuke Yamamoto), yang juga mampu memanfaatkan dan menggunakan Ghost Driver dan Eyecons untuk bisa dapat berubah menjadi Kamen Rider Specter (仮面ライダースペクター Kamen Raida Supekutā), dan juga Aran serta dermawan misterius lainnya yang dikatakan sebagai pemimpin misterius dari kumpulan monster-monster hantu bernama Ganma yang mereka kirim ke dimensi dunia manusia.
